Talc and anti-MUC1 antibodies.

چکیده
To the Editors: Cramer et al. (1) present very intriguing findings on anti-MUC1 antibodies and their relationship to ovarian cancer risk factors in healthy women, raising new theories on the role of immunity in ovarian cancer and opening up avenues for new research in this area. The analysis is straightforward, yet the conclusion about genital talc exposure increasing ovarian cancer risk via diminished antibody levels is not supported by their own data. Talc exposure was characterized as ‘‘none,’’ ‘‘body use only,’’ and ‘‘genital use.’’ Antibody levels were appropriately classified as ‘‘negative,’’ ‘‘low,’’ ‘‘intermediate,’’ and ‘‘high.’’ The overall unadjusted m P value is 0.01 but an analysis limited to none and body use only yields a strong association as well (P = 0.09). The genital use group had significantly lower intermediate levels of antibodies than the none group but an analysis of the body use only group also shows a significantly lower intermediate levels than the none group (P = 0.01). There is no biological expectation that using powder on the armpits or other skin surfaces above the perineum would affect MUC1 expression. Similarly, it seems highly unlikely that talc exposure on the superficial layers of female genitals could indirectly cause ovarian cancer by reducing antibodies. Talc from genital dusting could translocate to the ovaries but this speculative assumption was ruled out years ago by electron microscopy studies showing no relationship between genital dusting and ovarian talc particle concentrations (2).

Conditions associated with antibodies against the tumor-associated antigen MUC1 and their relationship to risk for ovarian cancer.
Many cancers, including ovarian, overexpress epithelial mucin (MUC1) and promote anti-MUC1 antibodies that may correlate with more favorable prognosis. By extension, risk for ovarian cancer might be reduced by preexisting MUC1-specific immunity. We measured anti-MUC1 antibodies in 705 control women, identified events predicting antibodies, and estimated ovarian cancer risk by comparing profiles...
متن کاملEffect of Anti-CD3/CD28 Dynabeads and Allogeneic PBMCs on Expansion of Anti-MUC1 Chimeric Receptor T Cells
Background and purpose: In recent years, immunotherapy using chimeric antigen receptor T cells (CAR T cells) has been considered as a novel and promising treatment for some diseases, especially cancer. The CAR T cell production is a multi-step, complex, time-consuming, and costly process. One of the most important steps in production of CAR T cells is expansion of these cells at appropriate num...
متن کاملSingle domain based bispecific antibody, Muc1-Bi-1, and its humanized form, Muc1-Bi-2, induce potent cancer cell killing in muc1 positive tumor cells
Muc1 is one of the most studied tumor antigens. However, antibodies or antibody-toxin conjugates against Muc1 have not shown significant efficacy for tumors with Muc1 overexpression. In this study, we employed bispecific antibody approach to target Muc1 positive tumor cells. A novel bispecific antibody, Muc1-Bi-1, was constructed by linking single domain antibodies, anti-Muc1-VHH and anti-CD16-...
متن کاملCell Surface-Associated Anti-MUC1-Derived Signal Peptide Antibodies: Implications for Cancer Diagnostics and Therapy
The MUC1 tumor associated antigen is highly expressed on a range of tumors. Its broad distribution on primary tumors and metastases renders it an attractive target for immunotherapy. After synthesis MUC1 is cleaved, yielding a large soluble extracellular alpha subunit containing the tandem repeats array (TRA) domain specifically bound, via non-covalent interaction, to a smaller beta subunit con...
متن کاملAntibody targeting of cell-bound MUC1 SEA domain kills tumor cells.
The cell-surface glycoprotein MUC1 is a particularly appealing target for antibody targeting, being selectively overexpressed in many types of cancers and a high proportion of cancer stem-like cells. However the occurrence of MUC1 cleavage, which leads to the release of the extracellular α subunit into the circulation where it can sequester many anti-MUC1 antibodies, renders the target problema...
